For detection of antibodies to the Erns protein of the classical swine fever virus (CSFV)

The pigtype CSFV Erns Ab is a double-antigen ELISA. The microtiter plate is coated with recombinant CSFV Erns antigen. During sample incubation, antibodies to the CSFV Erns protein bind to the immobilized antigen. Unbound material is removed by rinsing. Antibodies bound to the antigen are detected by a horseradish peroxidase (HRP) conjugate. Unbound conjugate is removed by rinsing. A colorimetric reaction is initiated by adding Substrate Solution and stopped after 10 minutes. If antibodies to the CSFV Erns protein are present in the sample, a blue color develops, which turns yellow after the addition of Stop Solution. The optical density (OD) is measured in a spectrophotometer at 450 nm. Measurement at a reference wavelength (620-650 nm) is optional. The OD value correlates to the concentration of the antibodies to the CSFV Erns protein in the sample.

酶標板上面包被了純化重組豬瘟Erns抗原。樣品經過孵育處理后，豬瘟Erns抗體能與包被的抗原特異性結合。洗板去除沒結合的物質后，每孔加入酶標記物孵育。洗板后，將直接使用的底物/顯色劑溶液加入各孔，如果待測樣品中含有Erns抗體，將發生藍色顏色反應。孵育完畢，加入終止液終止顏色反應并在酶標儀450nm測量。顏色反應的顯色強度與樣品中豬瘟Erns抗體含量成正比。
