a.由通道蛋白介導的擴散:這種擴散方式首先在哺乳動物的紅細胞中發現。通道蛋白貫穿膜,形成一個狹縫狀的中央親水通道,允許一定大小帶一定電荷的離子通過。像Cl- 即經此機制穿過紅細胞膜。這類蛋白存在廣泛,如細菌中的膜孔蛋白即屬此類。
b.由載體蛋白介導的擴散:這種擴散方式所涉及的載體蛋白是一類跨膜蛋白,它通過與物質結合而將物質運過膜。大多數物質,尤其是不溶于脂類的物質,即經這種方式運輸過膜。例如,葡萄糖即通過紅細胞膜上的一種特殊載體蛋白而被運入紅細胞內的。這種載體蛋白相當于結合在細胞膜上的酶,可同特異的物質結合,運輸過程中有類似于酶與底物作用的動力學曲線。
c.由離子載體介導的擴散:離子載體是溶于膜脂雙分子層的疏水性分子:它雖然也是按照被動轉運方式來轉運離子的,但是不同于載體蛋白,它包括載體性離子載體和通道形成性離子載體兩種。
纈氨霉素就是一種載體性離子載體,它在膜的一側結合K+,然后順著電化學梯度通過脂雙層,于膜的另一側釋放K+。大部分離子載體存在于微生物中,有的已被用作抗生素。
(2)主動運輸
物質經消耗能雖而被逆濃度梯度運輸通過生物膜的方式,即主動運輸。這是小分子物質和離子通過細胞膜的機制之二。其間所消耗的能量主要來自ATP。這種運輸方式也需要特定的蛋白載體。
① 離子泵
如生物膜上存在的Na+-K+泵、鈣泵、H+-K+泵、H+泵等,均屬此類。其中,Na+-K+泵是zui經典的子。Na+-K+泵,即Na+-K+ATP酶,是膜上的一種特殊蛋白。它利用水解ATP產生的能量,以逆離子濃度的方式向細胞外排出Na+,而同時將細胞外的K攝入細胞內。據計算,每消耗1個ATP分子可將3個Na+泵出細胞而將2個K+泵入細胞。
② 協同運輸
一種物質偶聯其他物質一起進行運輸的過程或方式叫協同運輸。它一般是間接利用ATP供能的。這種運輸方式另需要有關的ATP轉運離子以在膜的內外兩側建立離子濃度梯度,由此形成的電化學動力(或貯存的能量)才能使有關物質得以運輸過膜。
動物細胞中,葡萄糖和氨基酸等物質即經過質膜上的鈉泵和載體的協同作用才實現其逆濃度梯度的轉運。這里,載體蛋白與細胞外的Na+、葡萄糖(或氨基酸)等結合后,借助Na+-K+泵轉運Na+、K+時建立的電位梯度,將Na+、葡萄糖(或氨基酸)等同時運入細胞。而在細胞內從載體上卸下的Na+則又被Na+-K+泵運出細胞而維持Na+的電位梯度。
a. Channel-mediated Diffusion: This mode of diffusion is first found in mammalian erythrocytes. c. Diffusion mediated by ionophores: An ionophore is a hydrophobic molecule that is soluble in the lipid bilayer of a membrane lipid: although it transports ions in a passive transport mode, it differs from carrier proteins in that it includes a carrier ionophore and channel formation Two kinds of ionophores.
Valinomycin is a carrier ionophore that binds to K + on one side of the membrane and then passes through the lipid bilayer along an electrochemical gradient to release K + on the other side of the membrane. Most ionophores exist in microorganisms, and some have been used as antibiotics.
