第十五屆中國()檢驗(yàn)醫(yī)學(xué)暨輸血儀器試劑博覽會(huì)健侖體驗(yàn)

Hoechst是與DN特異結(jié)合的活性染料,儲(chǔ)存液用蒸餾水配成 mg/ml的濃度,使用時(shí)用PBS稀釋成終濃度為~mg/ml。DPI為半通透性,用于常規(guī)固定細(xì)胞的染色。儲(chǔ)存液用蒸餾水配 成mg/ml的濃度,使用終濃度一般為. ~mg/ml。結(jié)果評(píng)判:細(xì)胞凋亡過程中細(xì)胞核染色質(zhì)的形態(tài)學(xué)改變分為三期: Ⅰ期的細(xì)胞核呈波紋狀(rippled)或呈折縫樣(cresed),部 分染色質(zhì)出現(xiàn)濃縮狀態(tài);Ⅱ期細(xì)胞核的染色質(zhì)高度凝聚、邊緣化 ;Ⅱb期的細(xì)胞核裂解為碎塊,產(chǎn)生凋亡小體。.透射電子顯微鏡觀察結(jié)果評(píng)判:凋亡細(xì)胞體積變小,細(xì)胞質(zhì)濃縮。凋亡Ⅰ期(pro- poptosis nuclei)的細(xì)胞核內(nèi)染色質(zhì)高度盤繞,出現(xiàn)許多稱為 氣穴現(xiàn)象(cvittions)的空泡結(jié)構(gòu);Ⅱ期細(xì)胞核的染色質(zhì)高 度凝聚、邊緣化;細(xì)胞凋亡的晚期,細(xì)胞核裂解為碎塊,產(chǎn)生凋亡 小體細(xì)胞凋亡在胚胎發(fā)育、造血、免疫系統(tǒng)的成熟以及維護(hù)正常組織和 器官的細(xì)胞恒定與生長平衡,乃至機(jī)體衰老方面都起著重要作用。 因此,有關(guān)凋亡的研究在臨床和基礎(chǔ)等各個(gè)領(lǐng)域已經(jīng)廣泛開展,凋 亡細(xì)胞的檢測方法顯得非常重要。

Therefore, studies on apoptosis have been extensively carried out in clinical and basic fields, and detection methods for apoptosis have become very important.
